New real-time RT-PCR tests INSV and TSWV for plant material in horticultural crops
Publicationdate: 18 december 2023
We introduce the new real-time RT-PCR for the detection of INSV and TSWV in horticultural crops. Naktuinbouw Laboratories continues to support the sector by developing new tests to help diagnose plants with symptoms or to check for, and guarantee the absence of certain viruses.
The real-time RT-PCR is a technique for rapid amplification of RNA
This is done with the help of pathogen-specific building blocks. The amplification in RNA can be monitored digitally at any time. This makes it possible to confirm quicker and with more certainty whether the pathogen is present.
Viruses INSV and TSWV
- Impatiens necrotic spot virus (INSV) is transmitted by western flower thrips (Frankliniella occidentalis). Various ornamental crops can be infected, but vegetables like pepper. The virus can cause ringspots, but also necrosis. Depending on the crop species, it might stay local, only causing ringspots, in other crops the virus will move further into the plant.
- Tomato spotted wilt virus (TSWV) is also transmitted by western flower thrips. The host-plant range is broader than for INSV, occurring in many ornamentals and vegetable crops. Also, in these crops TSWV may stay local, or will become systemic.
Severe stem or top necrosis can be confused with bacterial infections. Both viruses can occur at the same time in one plant, although this is rare. INSV and TSWV are detected using one PCR, in which INSV and TSWV can be distinguished from each other.
