New seed extract PCR (SE-PCR) and culture method tests for crops

For the purpose of export requirements, the number of tests on different crops will be expanded. This concerns the following crops: eggplant, celery, hemp, watermelon, cucumber and pumpkin.

It concerns the following tests 

  • Solanum melongena (eggplant):  
    • Culture method for Clavibacter michiganensis subsp. michiganensis 
    • SE-PCR Clavibacter michiganensis subsp. michiganensis 
    • SE-PCR PSTVd, TCDVd and MPVd  
  • Apium graveolens (celery) 
    • SE-PCR Arabis mosaic virus (ArMV) 
    • SE-PCR Tobacco rattle virus (TRV)  
    • SE-PCR Tobacco streak Virus (TSV)  
  • Cannabis sativa (hemp): 
    • SE-PCR Arabis mosaic virus (ArMV) 
    • SE-PCR Tobacco rattle virus (TRV)  
    • SE-PCR Tobacco streak Virus (TSV)  
  • Citrulus lanatus, Cucumis sativus, Cucurbita maxima and Cucurbita pepo (watermelon, cucumber and pumpkin): 
    • SE-PCR Tobacco ringspot virus (TRSV)  
    • SE-PCR Tomato ringspot virus (ToRSV)

The plating method

The plating method is a technique used to isolate and identify bacteria. This is done using a specific culture medium. The pathogen will grow on this and can then be confirmed with a PCR. 

Real-time PCR technique

The (real-time) PCR is a technique for rapid amplification of DNA. This is done with the help of pathogen-specific building blocks. The increase in DNA can be monitored digitally at any time. This makes it possible to faster and with more certainty confirm whether the seed samples actually contain the pathogen.

To support the sector, Naktuinbouw Laboratories continues to develop new tests that can support diagnoses on plants and seeds with symptoms or just checking for and guaranteeing absence from certain plant pathogens.
